استكشاف التراكيب غير التقليدية لـ DNA في المناطق المحفزة للجينات المرتبطة بالسرطان ودورها في تنظيم التعبير الجيني

تتناول هذه الدراسة تحليل التراكيب الثانوية غير التقليدية من نوع G-quadruplex (G4) في المناطق المحفزة لعدد من الجينات المرتبطة بالسرطان ومقارنتها بجينات ضابطة منزلية، وذلك باستخدام أدوات المعلوماتية الحيوية. جرى استخراج تسلسلات المناطق المحفزة الممتدة 1000 زوج قاعدي قبل موقع بدء النسخ (TSS) لعشرة جينات مرتبطة بالسرطان، وهي MYC وTP53 وKRAS وEGFR وBCL2 وBRAF وPTEN وPIK3CA وNRAS وCDKN2A، إضافة إلى ستة جينات ضابطة منزلية، وهي GAPDH وACTB وHPRT1 وRPL13A وTBP وYWHAZ، من مرجع الجينوم البشري GRCh38/hg38 عبر UCSC Genome Browser، ثم حُللت باستخدام أداة QGRS Mapper للتنبؤ بتراكيب G4 وحساب قيم score-G الخاصة بها.
وأظهرت النتائج وجود اختلاف واضح بين المجموعتين من حيث عدد التراكيب المتوقعة وكثافتها وقوتها البنيوية، إذ بدت محفزات الجينات المرتبطة بالسرطان أكثر غنىً بتراكيب G4، مع تسجيل قيم أعلى لـ score-G مقارنة بالجينات الضابطة. كما لوحظ تمركز عدد كبير من هذه التراكيب بالقرب من TSS، ولا سيما في MYC وKRAS وNRAS وEGFR، بما يشير إلى احتمال وجود دور تنظيمي مباشر في ضبط التعبير الجيني. في المقابل، أظهرت الجينات الضابطة نمطًا أقل كثافة وأضعف بنيويًا وأكثر تشتتًا داخل المحفزات.
تدعم هذه النتائج الفرضية القائلة بأن تراكيب G4 في المحفزات قد تمثل عنصرًا تنظيميًا مهمًا في الجينات المرتبطة بالسرطان، وأن موقعها وقوتها البنيوية قد يرتبطان بآليات التحكم في النسخ. وبذلك، يمكن أن تسهم هذه التراكيب في فهم أعمق لتنظيم التعبير الجيني، كما قد تفتح المجال أمام دراسات تجريبية لاحقة لاستكشاف إمكاناتها كأهداف علاجية واعدة.
